SPECIFICATION

Organoids refer to tissue analogues with a certain spatial structure formed by three-dimensional cultivation of adult stem cells or pluripotent stem cells in vitro. Organs are highly consistent with their source tissues in terms of tissue structure, cell type, self-renewal ability, and function, thus demonstrating unique advantages in multiple biomedical fields such as developmental biology, disease modeling, precision medicine, drug research and development, gene and cell therapy, infection and immunity, and regenerative medicine.

Organoid Cryopreservation Medium (OCM) can be used for low-temperature long-term preservation of various mammalian organs and cell lines (such as humans, mice, pigs, bats, cows, etc.). This cryopreservation solution does not contain serum, any animal derived components, and contains 10% DMSO.

Product instructions
Freezing and Resuscitation of Organs (or Cells)
It is necessary to select organs (or cells) with good growth status for cryopreservation experiments.
1. Add pre cooled (2-8 ℃) Organoid Cryopreservation Medium to the recovered organoid (or cell), blow and mix well, and quickly transfer to the cell cryopreservation tube (the volume of cell suspension in the cryopreservation tube should be greater than 0.5mL).
Note: It is recommended to freeze 1x103-1x107 cells or corresponding organ like cells per 1mL of frozen solution.
2. Place the cryopreservation tube into the cell cryopreservation program cooling box (the cooling box must be balanced to room temperature or 4 ℃ in advance), and then immediately place the cooling box in an -80 ℃ ultra-low temperature refrigerator; The frozen storage tube can also be subjected to artificial gradient cooling treatment, such as standing at 4 ℃ for 10 minutes, storing at -20 ℃ for 1 hour, and storing at -80 ℃ overnight.
3. Transfer the frozen storage tube to liquid nitrogen for long-term storage under low temperature conditions (not higher than -70 ℃, recommended to use dry ice or original cooling box) the next day or 12 hours later.
Resuscitation of organoids (or cells)
4. Preheat the basic culture medium required for organs (or cells) at 37 ℃ in advance.
5. Quickly thaw the cell cryopreservation tube in a 37 ℃ water bath. When only a small amount of ice residue remains in the cryopreservation tube, immediately stop the water bath and transfer it to a clean operating platform in a timely manner.
6. Transfer the frozen suspension to a centrifuge tube, slowly add 5-10 times the volume of preheated basic culture medium, and gently mix well.
7. Centrifuge the organoid (or cell) suspension obtained in the previous step (horizontal centrifuge rotor, 150-300g, 3 minutes), discard the supernatant, and add basic culture medium again to resuspend the organoid (or cell) precipitation.
8. Centrifuge the suspension of organoids (or cells) obtained in the previous step (horizontal centrifuge rotor, 150-300g, 3 minutes). After discarding the supernatant, the organoids (or cells) obtained can be used for subsequent cultivation of organoids (or cells).
