SPECIFICATION

Host cell residual DNA (HCD) refers to DNA fragments from host cells that may appear in biological products. The residual DNA of host cells in biological products may transmit tumor or virus related genes, which are carcinogenic and infectious. The removal of impurities from host cells is a key step in the production of biopharmaceutical products. The detection of host DNA residues has become an important aspect of drug regulatory authorities' supervision of related biopharmaceutical enterprises. The DNA content in the final product should be as low as possible and needs to be determined using highly sensitive methods.
CHO DNA Assay Kit detection principle: The basic principle of Taq Man fluorescence probe method is to design and synthesize a probe that can specifically hybridize with the target gene. The 5 'end of the probe is labeled with a fluorescent group, and the 3' end is labeled with a quenching group. Under normal circumstances, the spatial distance between the two functional groups is very close, and fluorescent genes cannot emit fluorescence due to quenching. During PCR amplification, the primer and probe are simultaneously bound to the template, and the binding position of the probe is located between the upstream and downstream primers. When the amplification extends to the position where the probe binds, Taq enzyme utilizes 5 'exonuclease activity to cleave the fluorescent molecules connected to the 5' end of the probe from the probe, causing it to emit fluorescence. The number of detected fluorescent molecules is directly proportional to the number of PCR products, therefore, the initial number of DNA templates can be calculated based on the fluorescence intensity in the PCR reaction system.
