One tube RT-PCR is different from the traditional two-step method (reverse transcription and PCR amplification are completed in two steps). RNA template reverse transcription and cDNA amplification are completed in one tube, greatly reducing the risk of contamination, shortening analysis time, and improving the repeatability of the reaction. One Tube RT-PCR uses an optimized buffer system, high purity enzymes, and no DNA or RNA residues. High activity M-MLV reverse transcriptase (RNase H -) is used as the reverse transcriptase, and hot start high fidelity KOD enzyme is used for PCR amplification, significantly reducing mismatch.
RT-PCR products can be analyzed by agarose gel electrophoresis and EB staining, as well as downstream routine operations, such as restriction endonuclease digestion, ligation, DNA sequencing, dHPLC analysis; Alternatively, a quantitative PCR instrument can be used for qRT PCR for online analysis.
