This product contains ultra fast long segment DNA polymerase, dNTPs, and optimized reaction buffer at a concentration of 2 ×。 When using, simply add templates, primers, and replenish water until the final concentration of Mix is 1 × That's it. This product contains a red tracer dye and can be directly loaded for electrophoresis without the need for sample loading buffer; It can also be purified for subsequent operations such as enzyme digestion, ligation, and fluorescence sequencing.
Product features:
1. Ultra fast amplification speed: The extension speed can reach 5-10 seconds/kb, and the PCR can be completed in as little as half an hour.
2. Outstanding ability to amplify long fragments: capable of amplifying up to 10kb of long fragments (including complex templates such as the human genome).
3. Good fidelity: The fidelity is more than 3-6 times that of TAQ.
4. Strong amplification ability: High yield, with good results for complex templates with special structures and templates with high GC content.
Precautions:
1. The amplification speed of this system is extremely fast. Short fragments or simple templates such as plasmid templates can be extended at a speed of 5 seconds/kb and further shorten the PCR time by using fewer cycles. Long fragments (≥ 3kb) or complex templates with low production can use an extension speed of 15-20 seconds/kb or use a larger number of cycles.
When the yield of this product is low, it can be attempted to use longer extension times (such as 20 seconds/kb) and cycle numbers (such as 35) to increase the yield of PCR products.
For templates with high GC content, short segment pre denaturation and denaturation temperatures can be increased to 98 ℃ and/or the denaturation time can be appropriately extended. Long fragments should not increase the denaturation temperature to avoid DNA damage. The pre denaturation time should be appropriately extended to 5 minutes, and the denaturation time can be appropriately increased by 5-10 seconds.
If the GC content of the amplification template is high or the template is complex and the amplification effect is poor, DMSO can be added to the reaction mixture to the final concentration of 1% -8%, and the optimal concentration can be explored by increasing it in a 1% gradient. Alternatively, add betaine to the final concentration of 1-1.7M. And use Touchdown PCR
