Inspection principle:
After initial staining with crystal violet and iodine solution, a insoluble complex of crystal violet and iodine was formed in the cell wall. Gram positive bacteria, due to their thick cell wall, multiple layers of peptidoglycan network, and dense cross-linking, lost water during decolorization treatment with ethanol or acetone, resulting in reduced mesh size. In addition, it does not contain lipids, so ethanol treatment does not create gaps. Therefore, the crystal violet iodine complex can be firmly retained in the wall, Keep it purple; Gram negative bacteria, due to their thin cell wall, high lipid content in the outer membrane layer, thin peptidoglycan layer, and poor cross-linking degree, quickly dissolve the outer membrane mainly composed of lipids after encountering decolorizing agents. The thin and loose peptidoglycan network cannot prevent the dissolution of crystal violet and iodine complexes. Therefore, after decolorization with ethanol, they remain colorless, and then re stained with red dyes such as sand yellow, resulting in Gram negative bacteria showing red color.

Product Usage:
1. Fix the coating with flame, add (crystal violet solution) solution A to dye for 1 minute, and rinse off the dye solution with clean water.
2. Add (iodine solution) solution B to stain for 1 minute and wash with water.
3. Add (95% ethanol) solution C, shake the slide occasionally for about 10-30 seconds until there is no purple peeling, and wash with water.
4. Add (sand yellow re staining solution) solution D, dye for 1 minute, and wash with water.
5. Conduct oil mirror inspection after drying. Positive bacteria are purple, while negative bacteria are light red.
