Isolated Heart Perfusion System

No.BTK-PHA-001
This ex vivo cardiac perfusion system supports both Langendorff mode and Working Heart mode. It uses oxygenated Krebs-Henseleit or Tyrode’s solution to sustain isolated heart activity for cardiac physiology, pathology, pharmacology and myocardial metabolism research. The modular design allows upgrading from Langendorff to Working Heart configuration, with monitoring of pressure, ECG, myocardial tension and metabolic indicators for repeatable, stable experiments.

SPECIFICATION

Product Introduction
This system is a comprehensive set of equipment designed to perform ex vivo cardiac perfusion experiments. In these experiments, the heart is removed from an animal and connected to a specialized perfusion device. It is then perfused with a solution to maintain cardiac activity. This system is used to study changes in cardiac function and hemodynamics under pathological and physiological conditions. It is an effective tool for studying cardiac physiology, pathology, pharmacology, and myocardial biochemistry and metabolism. With its high repeatability and stability, this system ensures reliable research results.

Langendorff Perfusion System

  • Langendorff perfusion is a simple perfusion method in which oxygenated Krebs-Henseleit (KH) solution or Tyrode's solution is perfused through the aorta directly into the coronary arteries to maintain cardiac activity.
  • The perfusion solution does not enter the atria or ventricles because there is no fluid in the ventricles. This perfusion method is critical for demonstrating the mechanisms of cardiac pacing.
  • Typically, cardiac activity can be maintained for several hours. Parameters that can be monitored include perfusion pressure, ventricular pressure (using a balloon method), cardiac metabolism, electrocardiogram (ECG), myocardial tension, and others. This method also allows easy isolation of myocardium.

Working Heart Perfusion System

  • The Working Heart model, introduced by Neely et al. in 1967 as an adaptation of the Langendorff model, is widely adopted for its ability to better replicate physiological conditions. Its primary advantage lies in the adjustable left ventricular preload, allowing the heart to perform work similar to that in vivo. This setup closely approximates in vivo conditions when measuring cardiac energy metabolism and myocardial enzyme profiles.
  • In the Working Heart perfusion experiment, perfusate flows through the left atrium into the left ventricle. The ventricular contraction then overcomes aortic resistance, ejecting the solution into a simulated aortic pathway, thereby maintaining physiological perfusion conditions. Within this system, left ventricular filling pressure (preload) and aortic column height (afterload) can be held constant, accurately simulating in vivo cardiac workload. This approach provides significant insights for advanced research.
  • To enable researchers to customize systems according to their specific needs, this setup features a fully modular design. Starting from a Langendorff perfusion system, additional measurement components can be integrated to upgrade to a Working Heart perfusion model, allowing for flexible, tailored configurations.
