SPECIFICATION

This kit is based on LAMP technology and is used for qualitative detection of African swine fever virus in samples. The detection sensitivity is 10 copies. The main components of this kit are freeze-dried products, which can be transported at room temperature without affecting the performance of the kit.
LAMP is a novel nucleic acid amplification technique that uses 4 or 6 primers that can recognize 6 specific regions on the target gene. It relies on the strong strand displacement activity of Bst DNA polymerase and can amplify DNA 109-1010 times within 30-60 minutes. This reagent kit is a freeze-dried product, which is more stable, sensitive, and convenient to operate compared to liquid products. The detection is usually completed within 30 minutes.
This reagent kit is equipped with a color change reaction indicator, which contains hydroxynaphthol blue (HNB) indicator dye. The color of the positive amplification sample changes from violet to sky blue.

Usage:
1. LAMP HNB Reagent is in dry powder form, containing Bst DNA polymerase 4.0, dNTP, HNB colorants, etc. Before dissolution, the product can be stored for a long time at -20 ℃ and transported at room temperature without any degradation in performance. Before use, dissolve each bottle in 0.75ml of LAMP Buffer and use immediately. The remaining reagents can be stored at -20 ℃ for 1 month and stored for a long time below -60 ℃.
2. ASFV LAMP Primer is in the form of dry powder, which can be stored for a long time at -20 ℃ and transported at room temperature without any degradation in performance. After each bottle is dissolved in 0.2ml of ddH2O before use, it can be used immediately. The remaining reagents can be stored at -20 ℃ for 1 month and stored for a long time below -60 ℃.
3. The positive standard is in dry powder form, and 100% is added before first use μ Dissolve ddH2O in a vortex for 10 seconds and store at -20 ℃. The concentration of the dissolved product is 105 copies/ μ l. Use 1 for each experiment μ L is sufficient.
4. Prepare LAMP reaction mixture according to the following components

5. After the reaction system is prepared, place it at 60 ℃ for reaction for 30 minutes.
Attention: Do not open the reaction tube cover after the LAMP reaction is completed, otherwise it may cause aerosol pollution. Once aerosol pollution occurs, please use our company's aerosol DNA pollution remover (MT3001) for environmental cleaning.
6. Result interpretation
When the negative water control turns violet and the positive standard turns sky blue, the result interpretation is valid.
