SPECIFICATION
MTS is a new type of formazine compound, which belongs to the same tetrazole nitrogen derivative as MTT. MTS can be degraded by mitochondrial dehydrogenase in living cells to produce brown water-soluble formazane. By measuring the spectral absorption of formazane, the proliferation of cells can be measured. The principle is as follows: MTS can be degraded by mitochondrial dehydrogenase in living cells to produce a brown yellow water-soluble formamide principle
Product features:
1. This reagent kit is a single solution cell proliferation and cytotoxicity detection reagent, mainly consisting of MTS and an electron coupling reagent. The solution has strong stability and high sensitivity in the reaction.
2. Easy and fast operation. Can be directly added to the detection tablet. When tested in a 96 well plate, there is no need to wash or harvest cells, eliminating the dissolution step.
3. No radioactivity. There is no need to prepare liquid flash mixtures or dispose of waste.
Compared to MTT, it is safer to use. There is no need to use volatile organic solvents to dissolve formaldehyde products.
5. Flexible operation, different from MTT, the tablet can be placed back in the incubator for further incubation after reading to further generate color.
Storage conditions: Low temperature transportation, -20 ℃ dark storage, valid for six months.
Usage effect:Add 100 cells to the 96 well plate μ L/hole (approximately 1 × Incubate the cells in a 5% CO2 incubator at 37 ℃ for 24 hours, and add the appropriate concentration of the test compound. Incubate in the incubator for an appropriate time, adding 10 to each well μ MTS cell proliferation and toxicity detection solution for L. Incubate at 37 ℃ for 1-4 hours. Detect optical density at a wavelength of 490nm.
Memorandum:
1. Cell survival rate: Subtract the background OD value (complete culture medium with MTS cell proliferation and toxicity test solution, no cells) or the blank drug well OD value (complete culture medium with different dilutions of the test drug and MTS cell proliferation and toxicity test solution, no cells) from the OD values of each test well, and take the average OD value of each repeated well ± SD. The survival rate of cells is expressed in T/C%, where T is the OD value of the treated cells and C is the OD value of the control cells.
Cell survival rate%=(OD of treated cells/OD of control cells) × one hundred
2. Calculate the drug concentration (IC50) at T/C=50% and the drug concentration (IC90) at T/C=10%
