Verification principle
Peptone and glycerol provide carbon and nitrogen sources; Magnesium chloride and potassium sulfate promote the production of pseudomonas aeruginosa and fluorescein; Agar is a coagulant for culture medium.

Note: The culture medium contains a small amount of magnesium chloride, which may cause micro precipitation after sterilization.
Method and principle of pyocyanin production test:
Method: Select suspicious colonies and inoculate them on a culture medium (PDP) slant for measuring pyocyanin. Incubate for 24 hours and observe whether the slant has pigments. If there is any pigment, add 3.0-5.0mL of chloroform to the test tube and shake thoroughly to extract Pseudomonas aeruginosa from the culture in chloroform. When the chloroform extract appears green, add about 1mL of 1mol/L hydrochloric acid and let it stand for a while after shaking. If the upper hydrochloric acid solution appears pink to red, the test result is positive, otherwise it is negative.
Principle: Pseudomonas aeruginosa can produce pseudomonas aeruginosa, which can be extracted by chloroform and has a greater solubility in water than chloroform. It undergoes reversible redox reactions, with alkalinity appearing blue and acidity appearing red during oxidation. So after adding hydrochloric acid solution and shaking, pyocyanin will be transferred from chloroform to the hydrochloric acid solution and turn red in an acidic environment.

Prepare the culture medium according to the label usage, inoculate the following quality control strains, and incubate them aerobic at 36 ± 1 ℃ for 24 ± 2 hours.
