TRIS-EDTA antigen repair solution
EDTA antigen repair solution is a commonly used antigen repair solution that can be used for antigen repair of samples fixed with paraformaldehyde, formaldehyde, or other aldehyde reagents, such as paraffin sections and frozen sections. After cells or tissues are fixed with paraformaldehyde, formaldehyde, or other aldehyde reagents, many amino acid residues in the tissue form aldehyde or carboxymethyl bonds, blocking some antigenic determinants. At the same time, cross-linking between proteins also conceals antigenic determinants. This leads to weakened staining signals during immune staining and even some false positive staining results. Therefore, for paraffin sections, it is required to perform antigen repair before IHC staining, that is, to restore the original spatial form of the antigen by destroying the cross-linking formed between molecules during fixation. EDTA buffer is another commonly used IHC antigen repair solution in addition to citric acid buffer. Some antigens have better repair effects using EDTA buffer than citric acid buffer. Especially for certain nuclear antigens, the effect will be more pronounced.
This antigen repair solution uses widely used EDTA, which can effectively remove cross-linking between proteins caused by aldehyde fixing reagents, fully expose antigen epitopes in samples such as paraffin sections, and greatly improve the immune staining effect. Usually, paraffin sections require antigen repair treatment, while frozen sections may not undergo antigen repair treatment. Antigen repair can greatly improve the immunostaining effect of paraffin sections, but many literature studies have shown significant improvements in the staining effect of frozen sections. Especially when the immunostaining effect of frozen sections is not good, antigen repair can be attempted. From a theoretical perspective, whether frozen sections or cell slides, as long as the sample is fixed with paraformaldehyde, formaldehyde, or other aldehyde reagents for antigen repair, it will effectively remove the cross-linking between proteins, fully expose antigen epitopes, and greatly improve the immune staining effect.
